FDA ALERT [8/2007] This Alert highlights important revisions to the full prescribing information for pioglitazone HCl, marketed as Actos, Actoplus Met, and Duetact. The updated information includes a new BOXED WARNING, and additional updated WARNINGS, PRECAUTIONS and CONTRAINDICATIONS to emphasize that pioglitazone may cause or exacerbate heart failure, particularly in certain patient populations. The implications of this new labeling for healthcare professionals who prescribe Actos are summarized below. This information reflects FDA’s current analysis of data available to FDA concerning this drug. FDA intends to update this sheet when additional information or analyses become available.
New Warning for Actos
In September, the FDA launched a safety review of Actos after initial data from the manufacturer’s ongoing 10-year study suggested that the drug may increase the risk of bladder cancer.
The agency says it is also aware of a recent epidemiological study in France that also suggests an increased risk of bladder cancer associated with Actos. Based on this study, France has suspended use of the drug and advised not starting Actos in new patients.
Actos is part of a class of drugs known as thiazolidinediones that is used to treat type 2 diabetes. It is designed to help control blood sugar levels by increasing the body’s sensitivity to insulin.
The FDA says people currently taking Actos should continue taking it until advised otherwise by their health professional. Those who are concerned about the possible risk of bladder cancer should talk to their health care provider.

On June 15, 2011, the FDA issued an urgent health warning to users and prescribers of the drug Actos acknowledging the recent statistical data that shows up to a 40% increase in risk for bladder cancer in users of Actos. The studies further show that the risk of bladder cancer increases with higher dosages and longer courses of treatment with Actos
